Mounjaro Tirzepatide

Tirzepatide

WHAT IS IT?

Tirzepatide is a dual agonist that activates both the GLP-1 and GIP receptors. GIP (glucose-dependent insulinotropic polypeptide) is another hormone that affects appetite and metabolism.

HOW DOES IT WORK?

Tirzepatide binds to both GLP-1 and GIP receptors, leading to dual stimulation and an even more powerful effect on satiety and appetite reduction.

WHAT DOES IT TARGET?

  • Brain: This works on two receptors (GLP-1 and GIP) in your brain that help reduce your appetite. This results in stronger appetite suppression compared to medications that activate only one receptor.
  • Stomach and intestines: Slows down gastric emptying, providing a longer feeling of fullness after eating.
  • Pancreas: Activates two receptors in your pancreas to release insulin after eating, helping to keep your blood sugar levels stable. The primary effect is a powerful reduction in appetite.

BMI CLINIC BOOG

GENERAL INFORMATION

Mounjaro (tirzepatide) is a medication prescribed to adults with overweight (BMI of 27 or higher) or obesity (BMI of 30 or higher). It is used to aid in weight loss and is typically prescribed when weight loss through diet and exercise alone has not been sufficient. Mounjaro is intended to be used alongside a low-calorie diet and regular physical activity.

Mounjaro is administered once a week using a pre-filled injection pen. The injection is given subcutaneously (under the skin), typically in the abdomen, thigh, or upper arm. The pen is easy to use and does not require prior preparation.
Mounjaro contains a substance (tirzepatide) that mimics a natural hormone in your body. The medication helps increase the feeling of fullness and reduce appetite, which aids in reducing overall calorie intake.

Tirzepatide works by mimicking the hormones GLP-1 and GIP, which are naturally produced by the intestine in response to food. These hormones have several effects:

  • It suppresses appetite.
  • It slows down gastric emptying, making you feel full for longer.
  • It helps stabilize blood sugar levels, which is especially beneficial for people with type 2 diabetes.

These combined effects lead to a gradual reduction in calorie intake and support weight loss.

  • People using tirzepatide lost an average of 15-20% of their body weight within a year.

The treatment starts with a low dose of 2.5 mg per week and is gradually increased. The maximum maintenance dose is 15 mg per week. Gradual dose escalation is important to help the body adjust to the medication and to reduce potential side effects.

Mounjaro is not suitable for everyone. Do not use this medication if you:

  • Have a personal or family history of medullary thyroid carcinoma (MTC).
  • Have a condition known as multiple endocrine neoplasia syndrome type 2 (MEN 2).
  • Have severe liver or kidney problems.
  • Have previously had inflammation of the pancreas (pancreatitis).
  • Pregnant women or women who are breastfeeding.
  • Are allergic to tirzepatide or any of the other ingredients in Mounjaro.

Do not use it in combination with other weight loss medications or similar drugs.

Common side effects of Mounjaro include:

  • Nausea.
  • Vomiting.
  • Diarrhea.
  • Constipation.
  • Headache.
  • Fatigue.

Rarer but more serious side effects include:

  • Inflammation of the pancreas (pancreatitis).
  • Gallbladder problems.

Consult a doctor if you experience severe or persistent side effects.

  • Unopened injection pen: Store in the refrigerator (2°C to 8°C). Do not freeze.
  • Opened injection pen: Store in the refrigerator or at room temperature (up to 30°C) and use within 21 days.
  • Do not expose to extreme heat or freezing, as this may reduce its effectiveness.

For a comprehensive list of possible side effects, contraindications, and detailed information on usage, please refer to the official package leaflet. This can be accessed on the website of the Medicines Evaluation Board, via Apotheek.nl, or on the Mounjaro® website.

Mounjaro is injected subcutaneously once a week at a fixed time, preferably in the abdomen, thigh, or upper arm. The injection should not be administered into a muscle or blood vessel. Ensure that you use the injection pen correctly and store it properly according to the instructions in the package leaflet.

The effects of Mounjaro vary from person to person. Many individuals experience a reduction in appetite and initial weight loss after a few weeks. The greatest effects are often seen after several months, especially when the medication is combined with a diet and physical activity.

If you have missed a dose, take it as soon as possible, unless the next dose is within two days. In that case, skip the missed dose and continue with your normal schedule. Never take a double dose to make up for a missed one.
